Abstract

This paper presents a simple and rapid method for synthesizing monoesters and (un)symmetrical diesters using a rotary evaporator under reduced pressure, with diol or diacid as the starting material. The method involves a straightforward dehydrative esterification reaction between carboxylic acids and diols by efficient water removal using the rotary evaporator. This approach offers several advantages, including fast, high yield, ease of operation, and scalability, making it an attractive alternative to traditional esterification of diols/diacids. A series of monoesters and (un)symmetrical diesters were synthesized utilizing this protocol. The purity and structure of the synthesized products were verified through spectroscopic techniques. The versatility of the method allows for the synthesis of a diverse range of esters, providing a robust platform for the generation of prodrug candidates with tailored pharmacological profiles.
