Effect of cell membrane chlorine currents on energy metabolism of murine activated lymphocytes and bone marrow cells

Abstract

Chlorine channels have been found in immune cells but their functions were unclear. We have used MTT-assay to investigate the effect of CI channel blockers and Cl removal on energy metabolism (EM) of ConA-activated lymphocytes, EL-4 cells and bone marrow cells. The Cl channel blockers SITS and DIDS inhibited EM of ConA-blasts and EL-4 cells in a dose-dependent manner. Bone marrow cells were more resistant to inhibitory action of SITS and DIDS. Moreover, low and middle doses of SIRS resulted in some activation of EM of these cells. Besides, Cl channel blockers increased the level of intracellular Ca2+ in bone marrow cells. Our results suggest that the function of Cl channels in bone marrow precursors differs from that in more mature immune cells.
