The proposed project aims to develop a new DNA vaccine delivery system that can effectively express target proteins. The system involves displaying a cell-penetrating peptide on synthetic chromatin, which will be evaluated for its ability to deliver DNA into the nucleus of model human cells. To achieve this, recombinant proteins will be purified from E. coli, and the synthetic chromatin will be assembled using a self-assembly process. The functional characterization of the synthetic chromatin will be conducted by various cell-based assays with tissue culture and fluorescence microscopy technique.
